Osmoregulation of alkaline phosphatase synthesis in Escherichia coli K-12.

چکیده
Alkaline phosphatase, the phoA product, is synthesized constitutively in phoR mutants. This constitutive synthesis, which is independent of phosphate control, varies with changes in the osmolarity of the growth medium; phoA expression increases with increasing osmolarity. Maximum expression of the osmoregulated genes phoA, ompC, and ompF was achieved by osmotic manipulation of minimal medium; complex media repressed their expression.

The effect of amino acid analogues on alkaline phosphatase formation in Escherichia coli K-12. I. Substitution of triazolealanine for histidine.
In a histidine auxotroph of Escherichia coli K-12 that synthesizes alkaline phosphatase constitutively, replacement of histidine by 1,2,4-triazole-3-alanine causes an immediate inhibition of alkaline phosphatase production, but allows protein synthesis to continue for at least 40 min. Extracts of cells incubated with triazolealanine contain material that reacts with antibody spectic for the sub...

The substitution of canavanine for arginine in an arginine auxotroph of Escherichia coli K-12 allowed protein synthesis to continue but inhibited the formation of the alkaline phosphatase. Sonically disrupted extracts of cells incubated with 4C-canavanine contained 4C-protein that precipitated with antibody specific to alkaline phosphatase subunits and a fraction of 14 C-active enzyme that was ...
متن کاملSecretion of alkaline phosphatase subunits by spheroplasts of Escherichia coli.
Under conditions that permitted continued protein synthesis, spheroplasts of Escherichia coli were unable to form active alkaline phosphatase, although they synthesized protein that was antigenically related to alkaline phosphatase subunits. This cross-reacting protein was primarily detected in the medium of the spheroplast culture, and it had properties that closely resembled those of the alka...
متن کاملDerepression of alkaline phosphatase in Escherichia coli by p-fluorophenylalanine.
p-Fluorophenylalanine (FPA) causes a 100-fold increase in alkaline phosphatase in Escherichia coli B, strain PR1 at 30 C in minimal medium that contains excess inorganic phosphate (1.92 x 10(-3)m). Little increase in alkaline phosphatase synthesis occurs under these conditions at 22 C. [This strain is known to have a mutation in a regulator gene (R(2)) that, in the absence of FPA, permits derep...
متن کاملA dominant constitutive phoR mutation in Escherichia coli.
A dominant constitutive mutation of the phoR locus controlling alkaline phosphatase synthesis in Escherichia coli is described. Its phenotype can be explained by the production of a poisonous subunit of the phoR gene product. The phoR gene product is inferred to consist of at least 3 or 4 subunits.
